Beginning with the HGM8 workshop (1985), the location of IFNB3 on 2p23-qter has been considered confirmed (Povey et al., 1985). Todd and Naylor (1992) could not support this conclusion when they used specific sequence-based primers for PCR amplification of DNA sequences from microcell hybrid clones containing human chromosome 2. Therefore, they mapped the gene by analyzing a panel of somatic cell hybrids by PCR, using the same IFNB3 primer. This led them to the conclusion that the IFNB3 gene is located on human chromosome 8. Todd and Naylor (1992) suggested that, especially in mapping genetic sequences that belong to multigene families, PCR analysis has advantages over the standard Southern blot and in situ hybridization analyses; PCR is more sensitive, specific, and rapid.
