[1] Dipeptidyl carboxypeptidase I (DPKP-I) is one of the components of renin-angiotensin system (RAS), which is of critical importance in the maintenance of vascular homeostasis. This system includes <1> angiotensinogen (GEM:01q4/AGT) and products of its proteolysis, angiotensin I (A-I) and angiotensin II (A-II); <2> renin (angiotensinogenase; GEM:01q32/REN), which splits a decapeptide from the C-terminus of angiotensinogen to form A-I; <3> DPKP-I, which splits a carboxydepeptide from A-I to form A-II; <4> receptors for A-II, which genes are not yet specified.
[2] In systemic functional terms, cerebral, cardiac/renal (circulatory), and local tissue (intrcellular) RASs are distinguished. Molecular differences between tissue isoforms of DPKP-I have been found. Thus, a high-molecular isoform (160-170 kD; cDNA cloned: Soubrier-1988) primarily isolated from lung tissue and relatively low-molecular one (100-110 kD; cDNA cloned: Ehlers-1989), which is testicle-specific and androgen-dependent, were found. Both isoforms are encoded by the same DCP gene and the same pre-mRNA, being synthesized in different ways through alternative splicing, the testicle isoform (almost identical to the C-terminal part of lung isoform) appearing to be 30% shorter and to differ also in the N-terminal signal peptide (the first 36 amino acids)."
