TBX2 maps to Chr 17q23 in the human and Chr 11 in the mouse; presumably, a human TBX4 homolog maps to Chr 17q2 also (GEM:17q2/TBX4). TBX3 and TBX5 both map to human Chr 12 (GEM:12q241/TBX3; GEM:12q241/TBX5) and mouse Chr 5. Considering the close evolutionary relatedness of these 2 pairs of genes, a model of tandem duplication followed by clustered dispersion was proposed by Agulnik-1996 to account for the observed arrangement. If such a dispersion took place, it would be expected that other genes linked to the original cluster would be duplicated as well, and that one might find extended regions containing paralogous genes on mouse Chrs 5 and 11 (Ruvinsky-1997). Ruvinsky and Silver found 5 pairs of related genes in the mouse, in addition to the T-box clusters, that demonstrated a genomic distribution pattern consistent with the proposed en masse duplication event. The human homologs included NOS1 (GEM:12q242/NOS1) on Chr 12 and NOS2A (GEM:17q1/NOS2A) on Chr 17; and TCF1 (GEM:12q243/TCF1) on Chr 12 and TCF2 (GEM:17q1/TCF2) on Chr 17. Using data obtained from linked paralogous genes, Ruvinsky and Silver showed that the T-box cluster duplication occurred before the divergence between bony fish and tetrapods around 400 million years ago." |
