Gene: [08p22/CTSB] cathepsin B; amyloid precursor protein secretase;

FUN

The enzyme is a lysosomal proteinase.

FAG

The following human cathepsin genes and CTS-like sequences have been mapped: GEM:20q131/CTSA; GEM:08p22/CTSB; GEM:13q14/CTSBL; GEM:14q112/CTSG; GEM:11p155/CTSD; GEM:01q31/CTSE; GEM:15q2/CTSH; GEM:01q21/CTSK; GEM:09q22/CTSL; GEM:10^/CTSLL; GEM:10q/CTSLL1; GEM:10q22/CTSLL2; GEM:10q2/CTSLL3; and GEM:01q21/CTSS."

EVO

According to Bouyon-1989, the nucleotide sequence of the rat CTSB cDNA shares a 85% homology, with respect to the cathepsin region itself, with the human CTSB cDNA. However, the homology with respect to prepeptide, propeptide, or C-terminal region is only 50-70%. It is possible that in this case, as in the study of Murnane-1985, CTSB-like sequences or processed pseudogenes were found."

HIS

Data on the localization of the coding copy of CTSB are very controversial, although they are obtained by similar methods (blot hybridization of the genomic DNA from somatic hybrid cells with the gene-specific DNA probes, or in situ hybridization on chromosomes with the homologous probes). The locations reported are 11p13-11 (Murnane-1985); 8p22 (Wang-1987); and 13q14 (Bouyon-1989). The early data (Murnane-1985) on the location of a cluster of cathepsins B, H, and L in 11p13-11 (as indicated in the MIM catalogue) were not confirmed, although cathepsin D gene appears to be located in 11p15.5 ."
